In den vergangenen 20 Jahren ist ein stetig wachsendes Interesse im Bereich der Cannabisforschung zu verzeichnen. Grundlage dafür war die Entdeckung, dass Cannabinoide an spezifische Rezeptoren binden. Die sog. CB1-Rezeptoren werden in spezifischen Hirnarealen (z.B. Kleinhirn, Basalganglien und Hippokampus) und die CB2-Rezeptoren auf Zellen des Immunsystems exprimiert. Wenige Jahre nach der Entdeckung der Cannabinoidrezeptoren wurden auch körpereigene Liganden entdeckt (z.B. Anandamid). Diese Endocannabinoide wirken als Botenstoffe retrograd und regulieren die Erregbarkeit von Nervenzellen über eine CB1-Rezeptor abhängige Verminderung der Neurotransmitterfreisetzung. Über den CB2-Rezeptor, der auf Immunzellen exprimiert wird, wirken die Endocannabinoide als Immunmodulatoren. Es gibt eine Fülle von Untersuchungen, die nahe legen, dass eine Vielzahl physiologischer Prozesse mittels der beiden Cannabinoidrezeptoren moduliert werden, z.B. motorische Funktionen, Gedächtnis, Appetit, Schmerzwahrnehmung. Auch im Bereich der Suchtforschung wird das körpereigene Cannabinoidsystems zunehmend wichtiger.
In the past 20 years there is a growing interest in the field of cannabinoid-research. This is based on the discovery that cannabinoids act with specific receptors (CB1 and CB2). CB1-receptors are located in specific brain areas (e.g. cerebellum, basal ganglia, hippocampus) and CB2-receptors on cells of the immune system. Additionally, endogenous ligands of the cannabinoid receptors were discovered (e.g. Anandamide). These endogenous cannabinoids were proposed to be extracellular retrograde messengers, which regulates the excitability of neurons by CB1 receptor-dependent inhibition of neurotransmitter release. By means of CB2 receptors which are expressed on immune cells, the endogenous cannabionoid compounds act as immunmodulators. Evidence suggests that the two known cannabinoid-receptors are modulating many physiological processes: e.g. motor functions, memory, appetite, pain. Especially, in the field of addiction research the cannabinoid-system becomes more and more significant.
