Zusammenfassung
Die Entwicklung antiviral wirkender Substanzen kann rückblickend in drei Phasen unterteilt werden. Schon früh wurden Substanzen, die man ursprünglich als Tumoristatika einsetzte, daraufhin untersucht, ob sie auch die Virusvermehrung hemmen; hierzu gehörten Stoffe wie Ioddesoxyuridin oder Cytosinarabinosid. Ersteres war 1962 von Herbert E. Kaufman als eines der ersten antiviralen Therapeutika zur lokalen Behandlung der durch das Herpes-simplex-Virus induzierten Keratokonjunktivitis am Auge eingeführt worden. Mit dem fortschreitenden Wissen über die Molekularbiologie der Zelle und der Viren wurde jene überwiegend empirisch geprägte Vorgehensweise von Ansätzen abgelöst, mit denen man nur wenig selektiv wirkende Agenzien durch chemische Modifikationen zu verbessern suchte. Ergebnisse dieser „gezielten Empirie“ waren Substanzen wie Adenosinarabinosid, Bromvinyldesoxyuridin oder Acycloguanosin.
