Die Rolle von oxidativem Stress und Proteinglykierung

Zusammenfassung

Langsam ablaufende primäre Alterungsmechanismen führen zu anfangs unbemerkten Veränderungen im Gefäßsystem. Die endotheliale Dysfunktion ist ein früher Marker der Gefäßalterung. Ursache ist oxidativer Stress, der einerseits durch eine Reduktion der Bioverfügbarkeit von Stickstoffmonoxid (NO) und andererseits durch die Nitrosylierung wichtiger zellulärer Proteine vermittelt wird. Parallel dazu kommt es durch die Glykierung der extrazellulären Matrix zur Versteifung der Gefäße. In Kombination mit dem Verlust der elastischen Fasern wie Elastin führen diese Komponenten zu altersbedingten Gefäßveränderungen. Das Verständnis der Wirkung primärer Alterungsmechanismen könnte die Basis für die Entwicklung neuer Medikamente bilden.
